A blunt brash answer.
Optimus was not skilled with reading others, his observations only really holding any edge in this moment because it was Ratchet. Many years... COUNTLESS years however made him a bit sharper, latching onto the oddity firmly. Because of this he knew instantly that he must have hit the nail on the head if Ratchet's reply was to be taken as fact, a slight brow raise forming on his features to wordlessly convey that he was seeing through the defensive veil for what it most likely was. It helped that Ratchet didn't even make eye contact, glaring down at the screen to possibly hide any visual tells that couldn't be muddled under a gruff exterior. It read like a sneaky ploy to him, more so than the other mech keeping to task and working diligently.
Nothing was said as Ratchet continued, the medic's abrasive exterior flaring up to try to misdirect in some way and throw the situation aside as Optimus being unreasonable. The Prime simply watched through tired optics, his silence and expression saying more than words could to show that he most defiantly wasn't the more trusting and naive Orion any longer that would have backed down and trusted that Ratchet was fully correct. That said, back then Ratchet was not as roughened and beaten down from the War, so such a conversation would be completely different in days past, both adapting and growing a sharper edge that evolved through time up until this point.
It only took a bit of patience before his old friend seemed to lower the instant wall of bristled annoyance, speaking a bit more candidly. While he articulated his thoughts, Optimus eased back in his chair a bit while rolling his right shoulder slightly to try to stretch the cables and hydraulics that supported its movements around the hinge. It was an idle motion he didn't give much thought to, a sort of subconscious act.
"I appreciate you stepping outside your realm of expertise."
True on all counts. Walking into awkwardly unfamiliar territory can be intimidating in more ways than one. That said, it was only the two of them here, nobody else able to see any uncertainty or 'weakness' in either of them. It wasn't as if Optimus would tell anyone about any suggestions that were completely illogical, 'snitch' about something that was said that could be perceived as ridiculous. As of yet there wasn't anything that could remotely touch on such levels, so a lot of the frustration seemed unfounded to him.
"The views of an outsider untrained, can often provide a different perspective to kindle new actions."

Optimus liked sometimes throwing his Autobots into unfamiliar situations, as long as they were not dangerous. He would not pit an individual in a fight they were not comfortable in, yet he was not opposed to challenging with mental tasks to spin their processor. Either nothing changes, the Cybertronian finds a new way to look at things, or in very rare occasions they find an innate talent they never really considered before.
In a way he knew he was misdirecting his own slight breakdown and the focus that was on him onto Ratchet, but... it simply was what felt natural to him and occurred without second thought.
